Calcium-saturated calmodulin (Ca4-CaM) is involved in the regulation of a wide range of cellular functions. Dissociation of calcium from Ca4-CaM, and from complexes formed between Ca4-CaM and target peptides, frequently occurs in two kinetically distinct steps. The second (slowest) step, associated with the loss of two C-terminal calcium ions, can be studied by stopped-flow methods using a commercial spectrofluorimeter from Horiba JY equipped with an SFA-20/SPEX stopped-flow accessory.
